i am redoing my Cabinet for the Pi 4 T using Plywood, the present one i cut the port in the front but i think it would be nice if i can relocate it to the back or side of the speaker so that my son won't put his toys into the hole.

That will be just fine. There is no performance penalty from putting the port in the back, as long as it isn't obstructed. Angle the speaker a little from the back wall, giving the port room to breathe.
