I'm so close to finishing a pair of 4 Pi, but I glued the side piece of insulation in one speaker to the wrong side of the speaker, that is, the side opposite the port. All the other insulation is glued in place, including the piece across the brace. Will this make any difference? Do I need to pull it off and glue a piece to the correct side?

Go ahead and remove it and attach to the side nearest the port. It will peel away easily. Might leave a trace of fibers and glue lines - which might kind of "bug you" to know it's there - but that won't hurt anything and it'll stop bugging you in days. You're right, that wasn't bad at all. It probably does add a day for this to dry, though.
