Greetings from Poland to all forum members and a great thanks to Wayne for kindness, help and support.

Hopefully I will build my own 2Pi towers soon. I am curious, if different shaping of box and different speakers placement will critically affect sound? My ideas are as follows:

Any thoughts? I am especially interested in far right picture side design, as usually tweeter placement should be even to ears listening height...

I wouldn't do it because the position of the midwoofer and port is critical.  A cabinet this size and shape develops standing waves - transmission line behavior - in addition to Helmholtz (cavity) resonance.  So changing the position of the midwoofer will change the alignment of internal standing waves.

That's right.  The only one that has been modeled and measured during the design phase is what you show in picture one, the version that conforms to the plans.

I would expect that the second one might be OK but I haven't measured a speaker built that way so I can't say for sure.  I would expect the third and fourth pictures to be configurations that wouldn't work as well.  I would expect some ripple in the midbass from those because the standing waves along the vertical will line-up differently.
