Can the port be 6" wide by 3 1/2" tall and 10" deep? Or better yet, 10"w x 3 1/2" x 6" deep?

The port should be 6" x 3.5" and 10" long. It doesn't matter if the wide dimension is horizontal or vertical, although if the tweeter is mounted internally, it's a tight fit and has to go the way it is. But if you move the tweeter up on top and move the woofer up to keep the distances the same, you can change where the port is. Don't change the length though, it should always be 10". Also, remember this is for the stock box size. If you make a larger box, the port size changes. You want the Helmholtz frequency to be 38Hz.

Hope to have genuine 4 Pi Pro boxes glued up by Sunday night.

I'll put the horn on top and the woofer high up in the cabinet with the 6x3 1/2 slot below. Should be fun. Haven't built speakers in awhile.

I don't have the plans; center of horn to center of woofer should be 13 1/2"?

I'll send the plans. Get the woofer and tweeter as close as possible vertically.
