I know you don't want ports too small, but when there is a choice between one large diameter port, or two smaller diameter ports, is there any difference?

No difference if the Helmholtz frequency and total vent area are the same, provided individual port area isn't so small that air movement becomes turbulent.

My 18" reflex boxes have 8" dia ports and for a laugh my son (an adult(!)) put his head in one and pretended to be stuck. His friends thought it was funny and I regretted my wife taking the camera with her that morning.
