It really doesn't matter where you put the port usually. But the tower two π speakers are a little unusual in that they are very tall. Because of this, standing waves develop along the long dimension, so driver and port position become important. You don't want either the port or the driver to fall on a 1/4 node or odd multiples, as it would develop ripples in response. My suggestion is to leave the port where it is shown in the plans.
